Clobetasol

Clobetasol is a white to off-white crystalline powder with the chemical formula C22H28ClFO4, molecular weight 410.90700 and melting point 195~197℃. Clobetasol is a highly effective topical corticosteroid, mainly used for the treatment of chronic eczema, psoriasis, lichen planus, discoid lupus erythematosus, neurodermatitis, palmoplantar pustulosis and other skin diseases.

Dosage form
1. 0.05%ml.

Pharmacological effects
Clobetasol is a potent topical corticosteroid. It is readily absorbed from the skin by topical application, so it is particularly effective in the short-term treatment of persistent skin conditions. It has a strong capillary constricting effect and its anti-inflammatory effect is 112.5 times higher than that of hydrocortisone. It has no water and sodium retention effect, and has a certain effect of promoting sodium and potassium excretion. Clobetasol also has an inhibitory effect on DNA synthesis and mitosis of epidermal cells when applied topically, which is another mechanism for the efficacy of clobetasol in skin diseases characterized by cell renewal and proliferation, such as psoriasis. It also has antipruritic and vasoconstrictive effects, reducing spillage and inflammatory reactions.
Pharmacokinetics
Approximately 1% of clobetasol's therapeutic effect is transmitted through the skin when applied topically, and up to 10% of the drug is transmitted through the skin if the application site is covered with plastic film. The absorption of clobetasol can be increased by large areas, high concentrations or inflammation of the skin. In patients with hepatic insufficiency, the residence time of the drug in the body is prolonged accordingly.
Indications
For psoriasis, intractable eczema, lichen planus, discoid lupus erythematosus, etc. Clobetasol can be used when other hormones are not effective. It is used to treat skin inflammation and pruritus, such as neurodermatitis, contact dermatitis, seborrheic dermatitis, pruritus vulgaris, discoid lupus erythematosus, otitis externa, anal and vulvar pruritus, etc. .
Contraindications
1. Contraindicated in patients with hypersensitivity to clobetasol.
2. Early infectious skin lesions.
3. Skin ulcerative lesions.
4. Acne vulgaris, rosacea.
5. Perioral dermatitis.
6. Children.
Precautions
1. Clobetasol is prone to adverse reactions in pregnant women, children, face, axillae and groin and should be used with caution.
2. The initial dosage of clobetasol should be small. If severe irritation occurs, stop the drug immediately and take appropriate treatment. Clobetasol should not be used for various infections caused by fungi, bacteria and viruses.
3. Long-term use is not recommended to prevent the systemic effects of corticosteroids.
4. If skin irritation occurs when applying Clobetasol, discontinue use immediately and take appropriate measures.
5. Do not enter the eyes.
6. It should not be used for infectious skin diseases without the use of anti-infective drugs at the same time, and should be discontinued immediately if it is not effective within 1 week or if the infection spreads when used in combination with antibiotics.
7. Pregnant women, children (especially infants under 1 year old), and the face, armpits and groin should be prohibited.
8. Contraindicated in fungal, viral and bacterial infections, lupus erythematosus, acne and hypersensitivity to clobetasol.
