What’s Desloratadine powder?

Desloratadine (DESloratadine) is an active metabolite of loratadine obtained by deformating ethyl formate in vivo. It is a new third generation antihistamine with strong antiallergic effect, quick and lasting effect. It is a new drug for seasonal allergic rhinitis and chronic urticaria because of its affinity and selectivity to the terminal H1 receptor. Chemicalbook expressing human H1 receptors in vitro demonstrated 50 to 200 times the antagonistic activity of loratadine, fexofenadine or cetirizine, and it was safe and effective, well tolerated by patients, with a half-life of 21 to 24h, so only once a day. Desloratadine did not produce any effects related to QT interval and ventricular rate compared to terfenadine and asimidazole, both of which were banned from commercialization in 1999.

Function

1. Desloratadine has a strong anti-allergy effect. Desloratadine is highly selective and specifically binds to H1 receptors. It was 4 times stronger than loratadine in inhibiting histamine-induced foot swelling in mice. Desloratadine inhibits the release of inflammatory cytokines, including IL-3,IL-6,TNF-α and GM-CSF, from stimulated human mast cells and basophils at the namorl level, which is stronger than dexamethasone and cetirizine.

2. Desloratadine has the activity itself, good absorption, effective time less than 30 minutes. The maximum plasma concentration was reached about 3h after oral administration, the Cmax of single dose was 3.98ng·mL-1, and the clearance rate was 114 ~ 201L·h-1. A study of clinical onset time in patients with allergic rhinitis demonstrated that desloratadine was faster than existing antihistamines, with an average onset time of 28.45min, compared with 1h for loratadine and midazolastine and 2h for cetirizine. The half-life is about 24h and the effect lasts long.
