Lamivudine is a new antiviral drug, belonging to nucleoside reverse transcriptase inhibitors, which has strong inhibitory effect on hepatitis B virus (HBV) in vitro and experimental infected animals, and can inhibit the synthesis of HIV virus. The drug is made by GlaxoSmithKline. In the early 1990s, it was used to treat AIDS in some countries in Europe and North America. In the mid-1990s, medical experts found that it had an inhibitory effect on the DNA of hepatitis B virus. In 1998, the United States Food and Drug Administration (FDA) first approved Chemicalbook as a therapeutic drug for hepatitis B. China's State Food and Drug Administration approved the import of the drug, which was marketed in Chinese under the name Haputin, primarily as a treatment for hepatitis B and officially began selling it in mainland China in 1999. After 10 years of clinical trials, lamivudine is currently the only drug proven to delay the progression of hepatitis cirrhosis with fewer side effects and less cost.

Function

Lamivudine can be metabolized in HBV infected cells and normal cells to produce lamivudine triphosphate, which is the active form of lamivudine and is both an inhibitor of HBV polymerase and a substrate for this polymerase. The incorporation of lamivudine triphosphate into the viral DNA chain blocked the synthesis of viral DNA, and did not interfere with the metabolism of deoxyriboside in normal cells. The inhibitory effect on mammalian DNA polymerase α and β was weak, and the DNA content of mammalian Chemicalbook cells was almost not affected. It has no obvious toxicity to the structure, DNA content and function of mitochondria. Serum HBV-DNA assay of most hepatitis B patients showed that lamivudine rapidly inhibited HBV replication and its inhibition lasted throughout the treatment. At the same time, the serum transaminase decreased to normal, long-term application can significantly improve the inflammatory changes of liver necrosis and reduce or prevent the progression of liver fibrosis. Commonly used to treat chronic hepatitis B with hepatitis B virus replication.
