The Side Effect Of Ivermection

Ivermectin (ivermectin), also known as Aiwomil, is an effective drug against a variety of parasites in the abamectin family. It is suitable for the treatment of scabies caused by head lice and scabies mites, and onchocerciasis (river blindness), nematode infection, elephantiasis caused by bancroftia infection, etc. Ivermectin can be applied to the skin or taken by mouth, but contact with the eyes should be avoided.

 

 

Pharmacological effect

This product is a dihydro derivative of the macrolide avermectin Bla produced by Actinomycetes, which is a broad-spectrum antiparasitic drug. By binding to the glutamate-gated chloride ion channel (which is only expressed in invertebrates) in nerve cells and muscle cells in the worm, it causes parasite tonic paralysis and even death.

 

Pharmacokinetics

Peak plasma concentrations of ivermectin are reached 4 hours after oral administration. In adults, its terminal elimination half-life is about 27 hours, and the apparent volume of distribution is about 47L. About 93% is bound to plasma protein, and the tissue with the highest concentration is the liver and fat.

 

Indications

Ivermectin can be used for onchocerciasis, protozoa, scabies, etc.

 

Dosage

For the treatment of scabies, a single dose of 150-200 μg/Kg is taken orally, and the administration can be repeated after 14 days if necessary.

 

Side effect

Mazzoni reaction can occur during the treatment of onchocerciasis, which is an allergic reaction caused by the death of a large number of microfilariae;

When treating scabies, the side effects are mild, such as anorexia, weakness, headache, myalgia, arthralgia, fever, rash and itching;

It has been reported that the mortality rate of the elderly after taking ivermectin increased, but it is not necessarily related to ivermectin.

 

Precautions

1. It should not be used in combination with drugs that inhibit the central nervous system.

2. It should not be used in combination with P-glycoprotein inhibitors such as morphine and digoxin.

3. Patients with impaired blood-cerebrospinal fluid barrier such as African trypanosomiasis and meningitis are contraindicated.

4. Children younger than 5 years old or weighing less than 15kg should not be used.

5. The elderly, pregnant women, and lactating women should use it with caution.
